if you mean a complete tune (new set of strings etc.)
i lift up the floyd rose so it is horizontal (parallel with the fret board) and the remove the slack from the strings and start tuning.
you will need to tune each string about 4 times (or so i've found in the past.) but this should only be for a new set.
strings that have been tuned before, should only need adjusting at the BRIDGE if they are slightly out. (the round things that point in the air)
If when tuning your floyd rose, your bridge is not parallel with the guitar, then your sPrings wil need adjusting, which means taking the back off.
It takes me about 30-45 mins with a new set of strings (adjusting the bridge height and spring tension etc)
but a retune only takes 3-5 mins.
once you get it tuned right, it will save you a lot of hassle later on, if your springs are too loose, then the strings will be harder to retune, as they have more effect on the movement of the bridge.
hope something in that helps!!
