Don't hold me for my word, but I believe a riff is like the primary melody that you hear in a song. Take Day Tripper by the Beatles for example:
E:-----------------------------------------------|
B:-----------------------------------------------|
G:-----------------------------------------------|
D:----------2-0---4---0h2------Continues on------|
A:--------2-----2---2----------------------------|
E:0-0-3-4----------------------------------------|
That's the main melody that you hear in Day Tripper. This is considered a riff. The whole song is usually based on a single riff or riffs that the artists make up. Try making up some variations of the riff that fit well together and you've pretty much got the melody of your song. Usually riffs come from scales. So just take your base note and you can create a rythym from that.
Hope that helps.
