First you can look at the chord the song starts and/or ends with. It's not necessarily the song's key, but many times it is.
Then, look at the last two or three chords (cadences) of a phrase or the song. Many times it will end on (ii- or IV-) V-I. Other popular endings are I-V, IV-I, V-vi or even IV-V. On its own perhaps it's not worth much, but combine all these methods.
The best way surely is just to look the relationship of the chords in the song relative to each other. If a song uses A, f#m, D and E it's obvious that it's in A major (I, vi, IV and V) because it's the only key where those chords occur like that relative to each other.
As I said, combine these (and other) methods.
If you meant how do you *hear* it without knowing the chords: sorry, you'll need perfect pitch or some comparison.

If you know the chords it's easy to workout. If there are two major chords two frets apart those are the IV and V. If there is a dominant 7 chord then that is the V. If there are two minor chords two frets apart then they are the II and III.

To work out the chords if you don't know em just play along and experiment using your ear.

Perfect pitch is the ability to hear a tone and instantly know what pitch it is. Beethoven and Mozart had it, as does Yngwie Shredsteen.
